As is stated use the same fuel for running-in as you are planning to use normally, if you are normally using 25% use 25%, if you normally use 20% use 20%.
Of course you can change the % of nitro in the fuel, the only thing which happens if you normally use 25% nitro and you go to 20% nitro the engine will have less power.
Some persons run-in the engine with 10% nitro and when they finished the running-in they switch to 30% and race..., this is dangerous for the engine since using 10% nitro stresses less the engine then when using 30% if the engine is just run-in a problem can appear.
